纳米抗体源于在驼类和鱼类动物中发现的单重链抗体。与传统的IgG抗体不同,单重链抗体在构型上只 有重链而没有轻链。单重链抗体的可变区(VHH)被单独重组表达后,依然能够保持原本的生物学活性, 这种VHH即为我们常说的纳米抗体(Nanobody)。自上世纪80年代被发现后,纳米抗体一直吸引着人们 的目光,这是因为纳米抗体有着传统抗体不可媲美的优势,比如纳米抗体只有15Kd,容易穿透血脑屏障, 且在双抗构建中能完美替代成药性不高的scFv结构抗体;相对于传统抗体,纳米抗体稳定性高、特异性 高、同源性高,这些性质使其受到越来越多的关注。
